Описание

The dataset contains 438 records of Trichoptera species from 22 of the 23 families known from the Iberian Peninsula. Specimens were collected between 1975 to 2018 in Portugal, Spain and France (Paupério et al., 2023). Specimens have been identified to species or subspecies level, in a total of 141 species representing 37% of the Caddisflies known for the Iberian Peninsula. Specimens were captured during fieldwork directed specifically for the sampling of Trichoptera using different methodologies and stored in 96% ethanol. A tissue sample, usually a leg, was collected from each individual, from which DNA was extracted. Sequencing of the 658 bp COI DNA barcode was conducted within the InBIO Barcoding Initiative (IBI) and all DNA sequences were submitted to BOLD (Barcode of Life Data System) and GenBank databases. Specimens are deposited in the IBI collection at the CIBIO (Research Center in Biodiversity and Genetic Resources, Portugal) or in the collection Marcos A. González at the University of Santiago de Compostela (Spain). All DNA extracts are deposited in the IBI collection.

This dataset is composed of data relating to 438 Trichoptera specimens. All specimens were determined to species level except for two specimens that were identified to genus level. Overall, 141 species are represented in the dataset. Four species have been further identified to subspecies level: Wormaldia variegata mattheyi Schmid, 1952, Philopotamus montanus caurelensis González & Terra, 1979, Wormaldia variegata mattheyi Schmid, 1952, Rhyacophila dorsalis albarracina Malicky, 2002 and Rhyacophila mocsaryi tredosensis Schmid, 1952. The species represented in the dataset belong to 56 genera unevenly distributed, with the families Limnephilidae (11 genera, 19.6% of the total), Leptoceridae (8 genera, 14.3%), Glossosomatidae, Hydroptilidae and Psychomyiidae (each with 4 genera or 7.1% of the total) accounting for 55% of the genera represented in the dataset. Three families account for 43% of the species represented in the dataset, Limnephilidae and Rhyacophilidae (each with 23 species or 16.3% of the total) and Hydropsychidae (15 species, 10.6%).

Данные проекта

The InBIO Barcoding Initiative aims to develop a reference collection of DNA barcoding sequences covering Portuguese invertebrate taxa. Within the project, a special focus is afforded to insects, given their relevance to food webs, ecosystems functioning, high diversity and limited number of DNA barcoding sequences of interest.

Название The InBIO Barcoding Initiative Database: DNA barcodes of Iberian Trichoptera 01
Идентификатор InBIO Barcoding Initiative
Финансирование The InBIO Barcoding Initiative was funded by the European Union’s Horizon 2020 Research and Innovation Programme under grant agreement No 668981 and by the project PORBIOTA - Portuguese E-Infrastructure for Information and Research on Biodiversity (POCI-01-0145-FEDER-022127), supported by Operational Thematic Program for Competitiveness and Internationalization (POCI), under the PORTUGAL 2020 Partnership Agreement, through the European Regional Development Fund (FEDER). The fieldwork benefited from EDP Biodiversity Chair, the project “Promoção dos serviços de ecossistemas no Parque Natural Regional do Vale do Tua: Controlo de Pragas Agrícolas e Florestais por Morcegos” funded by the Agência de Desenvolvimento Regional do Vale do Tua, and includes research conducted at the Long Term Research Site of Baixo Sabor (LTER_EU_PT_002). SF was supported by an individual research contract (2020.03526.CEECIND) funded by FCT.
Описание района исследования Specimens were collected in nine continental Portugal districts, 16 Spanish provinces and one French department. One hundred and two different localities were sampled. Collection occurred between 1975 and 2018, although most specimens were collected in 2015 (25.4%), 2016 (17.8%) and 2017 (19.5%).
Описание плана выполнения исследований Specimens in this dataset were collected in field work carried out between 1975 and 2018, directed specifically for the sampling of Trichoptera using different methodologies. Specimens were morphologically identified and DNA barcoded.

Методы сбора

The studied material was collected in 142 different localities, 74 in Spain, 66 in Portugal and two in France. Sampling was conducted between 1975 and 2018, although 62% of the records were made between 2015 and 2017. Specimens were collected by direct search and individual netting of specimens, by sweeping the vegetation with a net or were directly collected at light traps, using both UV and mercury vapor lights. Captured specimens deposited in the IBI reference collection were preserved in 96% alcohol.

Охват исследования Portugal, Spain and France.
Контроль качества For all DNA barcoded specimens, their morphological identification was contrasted with the results of the Blast of the newly generated DNA barcodes in BOLD Identification Engine. Prior submission to GBIF, data was checked for errors and inconsistencies with OpenRefine 3.4 (http://openrefine.org/).

Описание этапа методики:

  1. Specimens were collected in 142 different localities in the Iberian Peninsula and France. Field work was carried out between 1975 and 2018, with 62% of the records made between the years 2015 and 2017.
  2. Specimens were collected during fieldwork by direct search and individual netting of specimens, by sweeping the vegetation or were directly collected at light traps (using both UV and mercury vapor lights) and preserved in 96% alcohol. Captured specimens were deposited in the IBI reference collection at CIBIO (Research Center in Biodiversity and Genetic Resources, Portugal) or in the collection Marcos A. González at the University of Santiago de Compostela (Spain).
  3. All specimens were morphologically identified using the available literature.
  4. DNA barcodes were sequenced from all specimens. For this, one leg was removed from each individual, DNA was then extracted, and a 658 bp COI DNA barcode fragment was amplified and sequenced. All DNA extracts were deposited in the IBI collection.
  5. All sequences in the dataset were submitted to BOLD and GenBank databases and to each sequenced specimen the morphological identification was contrasted with the results of the BLAST of the newly generated DNA barcodes in BOLD Identification Engine.
  6. Prior submission to GBIF, data was checked for errors and inconsistencies with OpenRefine 3.4 (http://openrefine.org/).
