Financement This work was funded by national funds through FCT – Foundation for Science and Technology under the PTDC/CTA-AMB/28511/2017 and DL57/2016/ICETA/EEC2018/25.
Description du domaine d'étude / de recherche Desertas subarchipelago is formed by three uninhabited small islets belonging to the Madeira Archipelago located 20 km southeast of Madeira Island. Deserta Grande is the largest of the three islets, with an area of approximately 10 km2 and a maximum altitude of 479 m. The Deserta Grande geomorphology is mostly rugged, with very steep slopes, ridges and peaks. The climate is temperate oceanic and the predominant habitats are rocky slopes and small arid flatlands, with sparse vegetation. Although fauna and flora of Desertas Islands are regarded as interesting and with high conservation value, freshwater biota was never study. Freshwater habitats are reduced to temporary streams in Vale da Castanheira.
Description du design Several freshwater habitats were sampled during two field campaigns in Deserta Grande, in 2013 and 2014. Diatom samples were collected by filtering water or by brushing the bottom and walls of the pools or stream bed. Samples were treated with warm nitric acid and mounted with Naphrax©. Diatoms slides were examined under 1000x magnification using a light microscope ZEISS AXIOIMAGE A1 with differential interference contrast. Species identification followed European diatom floras. At least 400 valves were counted in each sample.

Les personnes impliquées dans le projet:

Méthodes d'échantillonnage

Diatoms were sampled with help of a toothbrush to remove the biofilm and placed into a tray with a little water. The resulting suspension was collected in a plastic bottle, fixed with Lugol iodine and stored prior to analysis.

Etendue de l'étude Diatoms were sampled from several freshwater habitats were sampled during two field campaigns in Deserta Grande, in 2013 and 2014.
Contrôle qualité Samples for diatom analysis were following the National and European recommendations.

Description des étapes de la méthode:

  1. Diatom slides were mounted with Naphrax, and at least 400 valves per sample. were counted and identified to species or variety level under oil-immersion phase contrast light microscopy using a ZEISS AXIOIMAGE A1 microscope. General diatom floras were consulted for diatom identification.
