Descrição

The dataset contains 135 records of Hemiptera species collected from 2015 to 2019 in continental Portugal (Sousa et al., 2021). The species represented in the dataset, 90 in total, correspond to about 7.5% of the known true bugs diversity of continental Portugal, and contribute to the knowledge on the DNA barcodes and distribution of Portuguese Hemiptera. Specimens were captured during fieldwork by direct search of specimens or by sweeping the vegetation and stored in 96% ethanol. All specimens were identified to species level. A tissue sample, usually a leg, was collected from each individual, from which DNA was extracted. The DNA barcoding of these specimens was conducted within the InBIO Barcoding Initiative (IBI), funded by EnvMetaGen and PORBIOTA projects. DNA barcode sequences were deposited in BOLD (Barcode of Life Data System) online database. Preserved specimens and DNA extracts are deposited in the IBI collection at the CIBIO (Research Center in Biodiversity and Genetic Resources).

This dataset is composed of data relating to 135 Hemiptera specimens. All specimens were determined to species level or below. Overall, 90 species and one subspecies are represented in the dataset. These species belong to 31 families. The Pentatomidae family accounts for 20% of the total collected specimens, and no other family accounts for more than 8%. The Pentatomidae and Miridae families combined account for 26% of the total taxa represented, and no other family accounts for more than 7%. Eleven families are represented by a single taxon and ten by two taxa.

The InBIO Barcoding Initiative aims to develop a reference collection of DNA barcoding sequences covering Portuguese invertebrate taxa. Within the project, a special focus is afforded to insects, given their relevance to food webs, ecosystems functioning, high diversity and limited number of DNA barcoding sequences of interest.

Descrição da Área de Estudo Specimens were collected in 12 continental Portugal districts. Sixty different localities were, almost half of which (47%) from the Bragança district. Sampling was conducted from 2015 to 2019, with 39% of the specimens collected in the year 2018.

Métodos de Amostragem

The studied material was collected in 60 different localities from continental Portugal. Sampling was conducted between 2015 and 2019 on a wide range of habitats, both by direct search of specimens or using a hand-held sweep net. Collected specimens were examined using a binocular stereoscopic microscope and stored in 96% ethanol for downstream molecular analysis. Morphological identification was performed based on keys and descriptions from literature.

Área de Estudo Specimens were collected in 60 different localities of continental Portugal. Sampling was conducted from 2015 to 2019, in several field work expeditions.
Controle de Qualidade All DNA barcodes sequences were compared against the BOLD database and the 99 top hits were inspected in order to detect possible issues due to contaminations or misidentifications. Prior submission to GBIF, data was checked for errors and inconsistencies with OpenRefine 3.3 (http://openrefine.org).

Descrição dos passos do método:

  1. Specimens were collected in 60 different localities of continental Portugal. Fieldwork was carried out between 2015 and 2019.
  2. Specimens were collected during fieldwork by direct search of specimens or by sweeping the vegetation and preserved in 96% alcohol. Captured specimens were deposited in the IBI reference collection at CIBIO (Research Center in Biodiversity and Genetic Resources).
  3. Specimens were morphologically identified with the assistance of a stereoscopic microscope and using the available literature. A subset (23%) was identified using the BOLD Identification Engine directly.
  4. DNA barcodes were sequenced from all specimens. For this, one leg was removed from each individual, DNA was then extracted, and a 658 bp COI DNA barcode fragment was amplified and sequenced. For one specimens of Ceraleptus lividus only a 418 bp fragment was sequenced. DNA extracts were deposited in the IBI collection.
  5. All obtained sequences were submitted to BOLD and GenBank databases. The morphological identification of each sequenced specimen, when available, was contrasted with the results of the BLAST of the newly generated DNA barcodes in the BOLD Identification Engine.
  6. Prior submission to GBIF, data was checked for errors and inconsistencies with OpenRefine 3.3 (http://openrefine.org/).
