Descrição

The dataset contains two records of moth specimens (Lepidoptera, Depressariidae) collected in 2011 and 2015 in continental Portugal. The study of these specimens resulted in the description of a new Portuguese endemic species, Depressaria infernella Corley & Buchner, 2019 based on differences found in the male and female genitalia and DNA barcodes in Corley et al. (2019). Specimens were detected and captured using a mercury-vapor lamp to attract the insects. Captured specimens were identified to species level and preserved in 96% ethanol or in dry storage. From each specimen a tissue sample, one leg, was removed to be used for DNA extraction. The DNA barcoding of these specimens was conducted within the InBIO Barcoding Initiative (IBI), funded by EnvMetaGen and PORBIOTA projects. DNA barcode sequences were deposited in BOLD (Barcode of Life Data System) online database. Collected specimens and DNA extracts are deposited in the IBI collection at the CIBIO (Research Center in Biodiversity and Genetic Resources).

This dataset is composed of data relating to two Lepidoptera specimens. Both specimens were determined to species level. The dataset is composed of two species, Depressaria infernella Corley & Buchner, 2019 and Agonopterix scopariella (Heinemann, 1870). Both species belong to the family Depressariidae.

The InBIO Barcoding Initiative aims to develop a reference collection of DNA barcoding sequences covering Portuguese invertebrate taxa. Within the project, a special focus is afforded to insects, given their relevance to food webs, ecosystems functioning, high diversity and limited number of DNA barcoding sequences of interest.

Métodos de Amostragem

Specimens were captured in 2011 and 2015 in continental Portugal uisng mercury-vapor lamps as lures. Captured insects were preserved in 96% ethanol or in dry storage and stored at the InBIO Barcoding Initiative reference collection for downstream molecular analysis. Collected specimens were morphologically identified using a binocular stereoscopic microscope.

Controle de Qualidade The DNA barcoded specimen morphological identification was contrasted with the results of the Blast of the newly generated DNA barcode in BOLD Identification Engine. Prior submission to GBIF, data was checked for errors and inconsistencies with OpenRefine 3.3 (http://openrefine.org/).

Descrição dos passos do método:

  1. Specimens were captured in two localities of continental Portugal, in 2011 and 2015, using mercury-vapor lamps as lures.
  2. Captured specimens were preserved in 96% ethanol or in dry storage.
  3. The specimens were morphologically identified using a binocular stereoscopic microscope and resorting to available literature (Corley, Buchner & Ferreira, 2019; Hannemann, 1953; Palm, 1989.)
  4. A DNA barcode was sequenced from a tissue sample, one leg, that was removed. DNA was then extracted, and a 658 bp COI DNA barcode fragment was amplified and sequenced.
  5. The obtained sequences were submitted to BOLD database and the specimen morphological identification was contrasted with the results of the BLAST of the newly generated DNA barcode in BOLD Identification Engine.
  6. Prior submission to GBIF, data was checked for errors and inconsistencies with OpenRefine 3.3 (http://openrefine.org/).
