Description

The dataset contains three records of moth specimens of the family Gelechiidae collected from 2015 to 2018 in continental Portugal. The study of these specimens resulted in the description of a new genus, Mondeguina Corley & Rosete, 2020, a new species M. atlanticella Corley & Rosete, 2020, and a new genus placement for Catatinagma agenjoi (Gozmány, 1954) previously in the genus Apatetris Staudinger, 1880 (Corley et al. (2020)). The new genus contains M. mediterranella (Nel & Varenne, 2012) and M. atlanticella Corley & Rosete, 2020, a new Portuguese endemic species, described based on differences found in the male and female genitalia and on DNA barcodes. Specimens were detected and captured using mercury-vapor lamps to attract the insects. Captured specimens were identified to species level and preserved in 96% ethanol. A tissue sample, a leg, was collected from each individual, from which DNA was extracted. The DNA barcoding of these specimens was conducted within the InBIO Barcoding Initiative (IBI), funded by EnvMetaGen and PORBIOTA projects. DNA barcode sequences were deposited in BOLD (Barcode of Life Data System) online database. DNA extracts are deposited in the IBI collection at the CIBIO (Research Center in Biodiversity and Genetic Resources).

Méthodes d'échantillonnage

Sampling of specimens was done at three different localities of continental Portugal. Sampling was conducted between 2015 to 2018. Captured specimens were preserved in 96% and morphologically identified. DNA extracts are deposited in the InBIO Barcoding Initiative reference collection for downstream molecular analysis.

Contrôle qualité The DNA barcoded specimen morphological identification was contrasted with the results of the Blast of the newly generated DNA barcode in BOLD Identification Engine. Prior submission to GBIF, data was checked for errors and inconsistencies with OpenRefine 3.3 (http://openrefine.org/).

Description des étapes de la méthode:

  1. Sampling of specimens was done at three different localities of continental Portugal, between 2015 and 2018. Specimens were detected and captured at night by using mercury-vapor lamps as lures, although part of the specimens were collected a few metres away from the light, resting on grasses.
  2. The captured specimens were preserved in 96% ethanol.
  3. The specimens were morphologically identified using a binocular stereoscopic microscope and resorting to available literature (Corley, Rosete & Ferreira, 2017; Gozmány, 1954; Janse, 1949–1954, Junnilainen & Nupponen, 2010; Nel & Varenne, 2012; Sakamaki, 2000; Staudinger, 1880).
  4. DNA barcodes were sequenced from all specimens. For this, one leg was removed from each individual from which a 658 bp COI DNA barcode fragment was extracted, amplified and sequenced.
  5. The obtained sequences were submitted to BOLD database and the specimens’ morphological identification was contrasted with the results of the BLAST of the newly generated DNA barcode in BOLD Identification Engine.
  6. Prior submission to GBIF, data was checked for errors and inconsistencies with OpenRefine 3.3 (http://openrefine.org/).
