The dataset contains 13 records of Scorpionflies (Panorpidae) species collected from 2008 to 2019 in continental Portugal. All captured specimens were identified as Panorpa meridionalis, so far the only species of Mecoptera known to exist in Portugal Specimens were detected and captured by direct search of the environment. The dataset contains 13 records of Scorpionflies (Panorpidae) species collected from 2008 to 2019 in continental Portugal. All captured specimens were identified as Panorpa meridionalis, so far the only species of Mecoptera known to exist in Portugal Specimens were detected and captured by direct search of the environment. Sequencing of the 658 bp DNA barcodes was conducted within the InBIO Barcoding Initiative (IBI) and all DNA sequences were submitted to BOLD (Barcode of Life Data System) and GenBank databases.

The InBIO Barcoding Initiative aims to develop a reference collection of DNA barcoding sequences covering Portuguese invertebrate taxa. Within the project, a special focus is afforded to insects, given their relevance to food webs, ecosystems functioning, high diversity and limited number of DNA barcoding sequences of interest.

Specimens were captured from 2008 to 2019 in continental Portugal by direct search of ten sampling sites. Captured insects were preserved in 96% ethanol and stored at the InBIO Barcoding Initiative reference collection for downstream molecular analysis. Collected specimens were morphologically identified using a binocular stereoscopic microscope.

Охват исследования Specimens were collected in ten sites distributed among seven districts in the north of Portugal. Sampling was conducted from 2008 to 2019, in several field work expeditions.
Контроль качества To each specimen the morphological identification was contrasted with the results of the Blast of the newly generated DNA barcodes in BOLD Identification Engine. Prior submission to GBIF, data was checked for errors and inconsistencies with OpenRefine 3.2 (http://openrefine.org/).

Описание этапа методики:

  1. Specimens of Mecoptera were captured in continental Portugal from 2008 and 2019 by direct search of the sampling site. Collected specimens were preserved in 96% ethanol prior to storage.
  2. The specimens were morphologically identified resorting to available literature (Ward, 1983; Monserrat and Tillier, 2013).
  3. DNA barcodes were sequenced from 11 specimens. For this, one leg was removed from each individual from which a 658 bp COI DNA barcode fragment was extracted, amplified and sequenced.
  4. All obtained sequences were submitted to BOLD and GenBank databases and to each sequenced specimen the morphological identification was contrasted with the results of the BLAST of the newly generated DNA barcodes in BOLD Identification Engine.
  5. Prior submission to GBIF, data was checked for errors and inconsistencies with OpenRefine 3.2 (http://openrefine.org/).
