Descripción

The dataset contains 38 records of Blattodea collected from 2005 to 2019 in continental Portugal. Specimens were identified to species level, except for a single record identified to genus level. Of the eight species in the dataset, seven are cockroaches and one is a termite. Specimens were detected during fieldwork by direct search and stored in 96% ethanol. A tissue sample (one leg), was collected from each individual, from which DNA was extracted. The DNA barcoding of these specimens was conducted within the InBIO Barcoding Initiative (IBI), funded by EnvMetaGen and PORBIOTA projects. DNA sequences were deposited in the Barcode of Life Data System (BOLD) and GenBank databases. Preserved specimens and associated DNA extracts are deposited in the IBI collection at the CIBIO (Research Center in Biodiversity and Genetic Resources).

The InBIO Barcoding Initiative aims to develop a reference collection of DNA barcoding sequences covering Portuguese invertebrate taxa. Within the project, a special focus is afforded to insects, given their relevance to food webs, ecosystems functioning, high diversity and limited number of DNA barcoding sequences of interest.

Descripción del diseño Specimens in this dataset were collected in field work carried out between 2005 and 2019. The vast majority of the specimens were morphologically identified (80%) and DNA barcoded (75%).

Métodos de muestreo

The studied material was collected in 23 different localities from continental Portugal. Sampling was conducted between 2005 and 2019, with specimens collected by direct search during fieldwork and stored in 96% ethanol for downstream molecular analysis. Morphological identifications were performed using the available literature.

Control de Calidad All DNA barcodes sequences were compared against the BOLD database and the 99 top hits were inspected in order to detect possible issues due to contaminations or misidentifications. Prior submission to GBIF, data was checked for errors and inconsistencies with OpenRefine 3.3 (http://openrefine.org).

Descripción de la metodología paso a paso:

  1. Specimens were collected in 23 different localities of continental Portugal. Field work was carried out between 2005 and 2019, with 84% of the records made in the interval between 2015 to 2019.
  2. Specimens were collected during fieldwork by direct search and preserved in 96% alcohol. Captured specimens were deposited in the IBI reference collection at CIBIO (Research Center in Biodiversity and Genetic Resources).
  3. Specimens were morphologically identified with the assistance of a binocular stereoscopic microscope and using the available literature.
  4. DNA barcodes were sequenced from a subset of samples from each species in the dataset, selected to represent the best geographical coverage available for the species, for a total of 28 samples. For this, one leg was removed from each individual, DNA was then extracted, and a 658 bp COI DNA barcode fragment was amplified and sequenced. For two specimens of Reticulitermes grassei only a 325 bp fragment was sequenced. DNA extracts were deposited in the IBI collection.
  5. All obtained sequences were submitted to BOLD and GenBank databases and to each sequenced specimen the morphological identification was contrasted with the results of the BLAST of the newly generated DNA barcodes in BOLD Identification Engine.
  6. Prior submission to GBIF, data was checked for errors and inconsistencies with OpenRefine 3.3 (http://openrefine.org/).
