Description

The dataset contains 8 records of Snakeflies (Raphidioptera) species collected from 2016 to 2019 in continental Portugal. In total, three of the six species known to occur in Portugal are represented in the dataset. Specimens were detected and captured by direct search of the environment. Captured specimens were preserved in 96% ethanol. From each specimen one leg (tissue sample) was removed to be used for DNA extraction. Preserved specimens and DNA extracts are deposited in the InBIO Barcoding Initiative (IBI) collection at the CIBIO (Research Center in Biodiversity and Genetic Resources). Samples of each species were selected for DNA sequencing based on geographic provenance of the corresponding specimen. Sequencing of the 658 bp DNA barcodes was conducted within the InBIO Barcoding Initiative (IBI) and all DNA sequences were submitted to BOLD (Barcode of Life Data System) and GenBank databases.

The InBIO Barcoding Initiative aims to develop a reference collection of DNA barcoding sequences covering Portuguese invertebrate taxa. Within the project, a special focus is afforded to insects, given their relevance to food webs, ecosystems functioning, high diversity and limited number of DNA barcoding sequences of interest.

Méthodes d'échantillonnage

Specimens were captured from 2016 to 2019 in continental Portugal by direct search in six sampling sites. Captured insects were preserved in 96% ethanol and stored at the InBIO Barcoding Initiative reference collection for downstream molecular analysis. Collected male specimens were morphologically identified using a binocular stereoscopic microscope. Females specimens were identified using the BOLD (Barcode of Life Database) Identification System.

Etendue de l'étude Specimens of Raphidioptera were captured in six sites, distributed across two districts of the country. Sampling was conducted from 2016 to 2019, in several field work expeditions.
Contrôle qualité To each specimen the morphological identification was contrasted with the results of the Blast of the newly generated DNA barcodes in BOLD Identification Engine. Prior submission to GBIF, data was checked for errors and inconsistencies with OpenRefine 3.2 (http://openrefine.org/).

Description des étapes de la méthode:

  1. Specimens of Raphidioptera were captured in continental Portugal from 2016 and 2019 by direct search of the sampling site. Collected specimens were preserved in 96% ethanol prior to storage.
  2. The specimens were morphologically identified resorting to available literature (Monserrat and Papenberg, 2015). Females were identified using the BOLD (Barcode of Life Database) Identification System.
  3. DNA barcodes were obtained for all specimens. For this, one leg was removed from each individual from which a 658 bp COI DNA barcode fragment was extracted, amplified and sequenced.
  4. All obtained sequences were submitted to BOLD and GenBank databases and to each sequenced specimen the morphological identification was contrasted with the results of the BLAST of the newly generated DNA barcodes in BOLD Identification Engine.
  5. Prior submission to GBIF, data was checked for errors and inconsistencies with OpenRefine 3.2 (http://openrefine.org/).
