Descrição
The dataset contains 8 records of Snakeflies (Raphidioptera) species collected from 2016 to 2019 in continental Portugal. In total, three of the six species known to occur in Portugal are represented in the dataset. Specimens were detected and captured by direct search of the environment. Captured specimens were preserved in 96% ethanol. From each specimen one leg (tissue sample) was removed to be used for DNA extraction. Preserved specimens and DNA extracts are deposited in the InBIO Barcoding Initiative (IBI) collection at the CIBIO (Research Center in Biodiversity and Genetic Resources). Samples of each species were selected for DNA sequencing based on geographic provenance of the corresponding specimen. Sequencing of the 658 bp DNA barcodes was conducted within the InBIO Barcoding Initiative (IBI) and all DNA sequences were submitted to BOLD (Barcode of Life Data System) and GenBank databases.
This IPT archives the data and thus serves as the data repository. The data and resource metadata are available for download in the downloads section. The versions table lists other versions of the resource that have been made publicly available and allows tracking changes made to the resource over time.

Dados Sobre o Projeto
The InBIO Barcoding Initiative aims to develop a reference collection of DNA barcoding sequences covering Portuguese invertebrate taxa. Within the project, a special focus is afforded to insects, given their relevance to food webs, ecosystems functioning, high diversity and limited number of DNA barcoding sequences of interest.

Financiamento | InBIO Barcoding Initiative is funded by the European Union’s Horizon 2020 Research and Innovation programme under grant agreement No 668981 and by the project PORBIOTA — Portuguese E-Infrastructure for Information and Research on Biodiversity (POCI-01-0145-FEDER-022127), supported by Operational Thematic Program for Competitiveness and Internationalization (POCI), under the PORTUGAL 2020 Partnership Agreement, through the European Regional Development Fund (FEDER). The fieldwork benefited from EDP Biodiversity Chair, the FCT project PTDC/ASP-SIL/30250/2017 - POCI-01-0145-FEDER-030250 and includes research conducted at the Long Term Research Site of Baixo Sabor (LTER_EU_PT_002). |
Descrição da Área de Estudo | Specimens were captured during several field incursions from 2016 to 2019 in six localities, five in the district of Bragança and one in the district of Leiria. |
Descrição do Design | The specimens in this dataset were collected in field work incursions carried out between 2016 and 2019, encompassing several sites throughout. Specimens of Raphidioptera were detected by direct search. |
O pessoal envolvido no projeto:
Métodos de Amostragem
Specimens were captured from 2016 to 2019 in continental Portugal by direct search in six sampling sites. Captured insects were preserved in 96% ethanol and stored at the InBIO Barcoding Initiative reference collection for downstream molecular analysis. Collected male specimens were morphologically identified using a binocular stereoscopic microscope. Females specimens were identified using the BOLD (Barcode of Life Database) Identification System.
Área de Estudo | Specimens of Raphidioptera were captured in six sites, distributed across two districts of the country. Sampling was conducted from 2016 to 2019, in several field work expeditions. |
---|---|
Controle de Qualidade | To each specimen the morphological identification was contrasted with the results of the Blast of the newly generated DNA barcodes in BOLD Identification Engine. Prior submission to GBIF, data was checked for errors and inconsistencies with OpenRefine 3.2 (http://openrefine.org/). |
Descrição dos passos do método:
- Specimens of Raphidioptera were captured in continental Portugal from 2016 and 2019 by direct search of the sampling site. Collected specimens were preserved in 96% ethanol prior to storage.
- The specimens were morphologically identified resorting to available literature (Monserrat and Papenberg, 2015). Females were identified using the BOLD (Barcode of Life Database) Identification System.
- DNA barcodes were obtained for all specimens. For this, one leg was removed from each individual from which a 658 bp COI DNA barcode fragment was extracted, amplified and sequenced.
- All obtained sequences were submitted to BOLD and GenBank databases and to each sequenced specimen the morphological identification was contrasted with the results of the BLAST of the newly generated DNA barcodes in BOLD Identification Engine.
- Prior submission to GBIF, data was checked for errors and inconsistencies with OpenRefine 3.2 (http://openrefine.org/).
