Description

The dataset contains one record of the species Cochylimorpha punctiferana (Ragonot, 1881) (Lepidoptera, Tortricidae) collected in continental Portugal in 2016.The study of this specimen resulted in the resurrection of the Portuguese endemic species C. punctiferana from synonymy with C. discopunctana (Eversmann, 1844) based on clear differences in female genitalia and DNA barcode by Corley et al. (2020). A tissue sample, one leg, was removed to be used for DNA extraction. Sequencing of the 658 bp COI DNA barcode was conducted within the InBIO Barcoding Initiative (IBI) and the DNA sequence was submitted to BOLD (Barcode of Life Data System) and GenBank databases. The preserved specimen and DNA extract are deposited in the IBI collection at the CIBIO (Research Center in Biodiversity and Genetic Resources).

The InBIO Barcoding Initiative aims to develop a reference collection of DNA barcoding sequences covering Portuguese invertebrate taxa. Within the project, a special focus is afforded to insects, given their relevance to food webs, ecosystems functioning, high diversity and limited number of DNA barcoding sequences of interest.

Sampling Methods

The Cochylimorpha punctiferana specimen was captured in continental Portugal in 2016. The captured specimen was preserved in 96% ethanol and stored at the InBIO Barcoding Initiative reference collection for downstream molecular analysis. The collected specimen was morphologically identified.

Study Extent A single specimen was collected in Vale da Porca, in Continental Portugal, in 2016.
Quality Control The DNA barcoded specimen morphological identification was contrasted with the results of the Blast of the newly generated DNA barcode in BOLD Identification Engine. Prior submission to GBIF, data was checked for errors and inconsistencies with OpenRefine 3.3 (http://openrefine.org/).

Method step description:

  1. The Cochylimorpha punctiferana specimen was captured in a single locality of continental Portugal, in 2016.
  2. The captured specimen was preserved in 96% ethanol.
  3. The specimen was morphologically identified using a binocular stereoscopic microscope and resorting to available literature (CORLEY & FERREIRA, 2020; KENNEL, 1908-1921, 1913; RAGONOT, 1881; RAZOWSKI, 1970; RAZOWSKI, 2002, 2009).
  4. A DNA barcode was sequenced from a tissue sample, one leg, that was removed. DNA was then extracted, and a 658 bp COI DNA barcode fragment was amplified and sequenced.
  5. The obtained sequence was submitted to BOLD and GenBank databases and the specimen morphological identification was contrasted with the results of the BLAST of the newly generated DNA barcode in BOLD Identification Engine.
  6. Prior submission to GBIF, data was checked for errors and inconsistencies with OpenRefine 3.3 (http://openrefine.org/).
